My grandmother recently passed away and we would like to root her Peonies. Is this possible to do?

Cuttings are very difficult, if not impossible, alas...They are all propagated by grafting, or by divisions. If you're talking about herbaceous peonies, the best (and nearly only) way is to dig a division of them (2 or 3 stems, not more, or if you dig more, divide them into smaller divisions with 2 or 3 stems before replanting). As for tree peonies, it's even worse. Root cuttings are supposed to be possible, but very hard, especially at this season. Some of them, though, like 'Duchesse de Morny', tend to have several branches starting from the rootstock, and can be divided (I've just done it with my late mother's DDM peony).

Thank you! I had a feeling I would need to divide it. Is there a best time to do this? Fall? or is summer division okay?

According to my books, division is best performed when the plant has stopped its vegetation cycle, i.e. between mid-august and mid-march.
